well for the past six months I've been using a honking big messenger bag for a purse so quite a lot. It varies from week to week but the constants are:
* Keys (to my own apartment, my car, work, etc.)
* 2 year planner
* Wallet (which is actually a zip top change purse with my various cards in a cardholder)
* Chapstick
* Cell phone
* swatches of Dunlop tartan, my wedding dress and trim, and the bridesmaid dresses fabric
* pocket pack of tissues
* pens, including a Red Dwarf one
* prescription sunglasses in a hard case
* umbrella (which has been in there for about a month now)
Two year planner?? Well, I guess with the wedding coming up, it sort of makes sense you'd want the messenger bag with the two year planner and the swatches.
My Red Dwarf pen has run out - I'm very bummed that there doesn't seem to be any way to even open it to refill it. :( I was keeping it in my pocket at work - because one really needs a pen, all the time, at work - and it provided lots of conversation.
My wallet. It's no Costanza wallet. In fact, it's the antithesis of that... very slim. It has some cash (occasionally), my driver's license, debit card, and CTA pass.

* Wallet, containing driver's license, bank card, a bunch of "buy a bunch, get one free" cards from local businesses, change, pictures of my nieces, and (occasionally) some cash.

You carry around a checkbook! You're one of a dying breed since the advent of debit cards, I think. I only use mine for paying bills, so it stayes home.
Oh, and my keychain. It's just one of the hiking clip dealies with the front door key, back door key, mail key, bike key, and the clicker for the front door at work.
* Keys to my car, Aaron's truck, my post office box, my parents house, and my house. On a Red Sox key ring, of course.
* My wallet, which other than money contains: My license, debit card, company credit card, health/dental/prescription insurance cards, AAA card, library card, Bull Moose frequent buyer card, Avis Preferred card, Marriott Rewards card, a Home Depot gift card, and my membership cards for Planned Parenthood and NOW. I need to find my ACLU card so I can really, really annoy my father with it. ;)
* A calculator
* A hairbrush and an elastic
* Burt's Bees lip balm
* A packet of Emergen-C
* A lighter
* Cigarettes
* My cell phone
* My iPod.
If I want to not carry my purse, it's reduced to keys, debit card/license/cash stuffed in my back pocket, cigarettes, lighter, and cell phone.
Also, unrelatedly, I just had to walk up to the bar I was at last night to get my car. Now I am dying from the heat. I am a wimp.
Wow, you've got what I call (when I have to search them at work) a desk-purse - everything I leave on my desk is in your purse. Well, not everything (she said, eyeing the pile on her desk).

-Wallet (green embroidered silk containing ID, bank and library card, occassional monies, a list of the things my friend Mellie loves about me, and a tiny diploma from 'The Martin A. Buzas School of Charm and Tact that I got as a joke on finishing my first internship with the Smithsonian).
-Sunglasses
-Cell phone (on silent as I don't like being subjected to trying to hold a conversation and navigate a sidewalk at the same time).
-Metro farecard
-Two pennies and a Maryland state quarter.
-Organic lipbalm and lotion.
-One brown marble fountain pen (Levenger's TrueWriter) filled with brown ink and inscribed with my initials (EYH), a gift from my father for my birthday.
Does the pen remind you to write? I carry around a silver Cross pen my father gave me, almost as a talisman to remind me to write (stories, letters, journal, anything at all, just write).
It sure does! I always try to carry around a little bit of paper, too, so that I can note things as they happen (even if most of what I do with that is just for LJ-ing purposes).
